In order of most used screensizes that visit my site:

60.03% of visitors use 1024x768
20.65% of visitors use 1280x1024
8.78% of visitors use 800x600

Best way to tell is to setup Google Analytics and it will tell you what resolutions your visitors are using.

according to Google analytics my users screen sizes are

1024x768 - 61%
800x600 - 13%
1280x1024 - 6%
1280x800 - 9%

My website layout doesn't fit the popular 1024x768 resolution, it stretches the screen a little bit, I need to get around to fixing that up lol.
